ABA2 encodes a cytosolic short-chain dehydrogenase/reductase that catalyzes the NAD-dependent conversion of xanthoxin to abscisic aldehyde, a key late step in the primary abscisic acid (ABA) biosynthetic pathway derived from carotenoids. Positioned downstream of NCED-mediated 9-cis-epoxycarotenoid cleavage and upstream of AAO3 (abscisic aldehyde oxidase), ABA2 is rate-influential for ABA accumulation in response to drought, salinity, and developmental cues such as seed maturation. Loss-of-function mutations in ABA2 reduce ABA levels, leading to impaired stomatal closure, enhanced transpirational water loss, reduced seed dormancy and desiccation tolerance, and altered stress-responsive gene expression; conversely, overexpression can elevate ABA content and stress resilience. ABA2 is broadly expressed with enrichment in vascular and guard cell-associated tissues and is transcriptionally and post-transcriptionally regulated by environmental signals and metabolic status. As a central node linking carotenoid metabolism to ABA signaling outputs, ABA2 impacts plant water-use efficiency, growth-stress tradeoffs, and seed physiology.
