Western blot analysis was performed on whole cell extracts (30 µg lysate) of A549 (Lane 1), COS-7 (Lane 2), MDCK (Lane 3), HT-29 (Lane 4), C2C12 (Lane 5), PC-12 (Lane 6), U-2 OS (Lane 7) and tissue extract (30 µg lysate) of Rat Testis (Lane 8), Mouse Liver (Lane 9). GAPDH Antibody — FIG 1 . RNAi-based assay to identify host factors required for intracellular survival of Y. pestis . To determine whether reproducible RNAi could be achieved in RAW264.7 macrophages, cells were reverse transfected with siRNAs targeting indicated genes. (A and B) Forty-eight hours posttransfection, cells ( n = 3) were harvested for RNA isolation and qRT-PCR (data represent the level of gene expression compared to the level for the scrambled siRNA control) (A) or protein isolation for Western blot analysis (beta-actin was used as a loading control) (B). alpha-GAPDH, anti-GAPDH antibody; alpha-betaactin, anti-betaactin antibody. To demonstrate that the Y. pestis CO92 pCD1 (-) Lux PtolC bioreporter accurately represents intracellular bacterial numbers, RAW264.7 macrophages were infected with Y. pestis CO92 pCD1 (-) Lux PtolC at the indicated MOIs ( n = 12), and extracellular bacteria were killed with gentamicin. (C) Bioluminescence (in relative light units [RLU]) of intracellular bacteria was determined at 1, 4, 8, and 18 h postinfection. PA1-988 detects rat GAPDH in human HeLa cell lysates and mouse and rat tissues..

PA1-988 has been used successfully in Western blotting. By Western blot this antibody specifically detects a ~36 kDa protein representing GAPDH.

PA1-988 immunizing peptide corresponds to amino acid residues 4-18 from human GAPDH.

GAPDH (Glyceraldehyde-3-phosphate dehydrogenase) is a catalytic enzyme commonly known to be involved in glycolysis. GAPDH exists as a tetramer of identical 37-kDa subunits and catalyzes the reversible reduction of 1,3-bisphosphoglycerate to glyceraldehyde 3-phosphophate in the presence of NADPH. Apart from playing a key role in glycolysis, GAPDH is ubiquitously expressed and displays other activities unrelated to its glycolytic function. GAPDH is reported to be involved in the processes of DNA replication, DNA repair, nuclear RNA export, membrane fusion and microtubule bundling. Studies provide evidence of GAPDH playing an essential part in gene expression observed in apoptosis and as part of the cellular phenotype of age-related neurodegenerative diseases. Further, GAPDH is involved in other cellular processes ranging from membrane fusion, and neuronal apoptosis in cancer. GAPDH is reported to bind to a variety of other proteins, including the amyloid precursor protein, mutations in which cause some forms of Alzheimer's disease (AD), and the polyglutamine tracts of Huntingtin, the protein product aberrant forms of which are causative of Huntington's disease. Associations between GAPDH, actin and tubulin have also be reported. Since GAPDH is expressed at high levels in most tissues, it is useful as protein loading control in Western Blot analysis.
