The LRRC4 gene, also known as netrin-G ligand 2 (NGL-2), is a brain-specific member of the leucine-rich repeat superfamily, playing a critical role in the central nervous system. Its expression is particularly high in areas such as the hippocampal CA1 and CA2 regions, as well as in granule cells of the dentate gyrus. LRRC4 is a known tumor suppressor gene associated with glioma suppression, as it regulates glioma cell growth, angiogenesis, and invasion through extensive signaling networks. It can participate in multiphase loops involving miRNA, transcription factors, and gene methylation modifications. This gene targets various genes involved in cell motility, impacting glioblastoma cell movement and polarization by binding to enhancer elements and activating transcription, notably the RAP1GAP gene. Its function and expression are crucial in maintaining normal brain function and hold potential implications in the management of gliomagenesis.
