P2X7 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of P2X7 in paraffin-embedded esophageal cancer tissue. Sample was incubated with P2X7 monoclonal antibody (Product # MA5-31813) using a dilution of 1:200-1:1000 followed by DAB staining.

P2X7 Antibody in Western Blot (WB) — Western Blot was performed using Anti-P2X7 Monoclonal Antibody (7G1D6) (Product # MA5-31813) and a 59 kDa band corresponding to P2X purinoceptor 7 was observed across cell lines and tissue. Membrane enriched extracts (60 µg lysate) of U-937 (Lane 1), SH-SY5Y (Lane 2), A-375 (Lane 3), B16-F10 (Lane 4), RT-4 (Lane 5), K-562 (Lane 6), DU 145 (Lane 7), Mouse Brain (Lane 8), Mouse Skeletal Muscle (Lane 9), Rat Brain (Lane 10), Rat Skeletal Muscle (Lane 11) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # LC2001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).

P2X7 Antibody in Western Blot (WB) — Western blot analysis of P2X7 in A431 (1), U251 (2), HeLa (3), U937 (4), and HepG2 (5) cell lysate. Samples were incubated with P2X7 monoclonal antibody (Product # MA5-31813) using a dilution of 1:500-1:2000.

P2X7 Antibody in Western Blot (WB) — Knockdown of P2X purinoceptor 7 was achieved by transfecting A-375 with P2X purinoceptor 7 specific siRNAs (Silencer® select Product # S9959, S9961). Western Blot analysis (Fig. a) was performed using Membrane enriched extracts from the P2X purinoceptor 7 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with P2X7 Monoclonal Antibody (7G1D6) (Product # MA5-31813, 1:1000 ) and Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:20000). Densitometric analysis of this Western Blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to P2X purinoceptor 7.

P2X7 Antibody in Flow Cytometry (Flow) — Flow cytometry of P2X7 in HeLa cells (green). Sample was incubated with P2X7 monoclonal antibody (Product # MA5-31813) using a dilution of 1:200-1:400 followed by negative control (red).

P2X7 Antibody in ELISA (ELISA) — ELISA analysis of P2X7 in Control Antigen (black line, 100 ng); Antigen (purple line, 10 ng); Antigen (blue line, 50 ng); Antigen (red line, 100 ng). Samples were incubated with P2X7 monoclonal antibody (Product # MA5-31813) using a dilution of 1:10,000.

The product of this gene belongs to the family of purinoceptors for ATP. This receptor functions as a ligand-gated ion channel and is responsible for ATP-dependent lysis of macrophages through the formation of membrane pores permeable to large molecules. Activation of this nuclear receptor by ATP in the cytoplasm may be a mechanism by which cellular activity can be coupled to changes in gene expression. Multiple alternatively spliced variants have been identified, most of which fit nonsense-mediated decay criteria.
