PLCG1 Antibody (PA5-85807) in ICC/IF
Immunofluorescence analysis of PLCG1 was performed using 70% confluent log phase NTERA-2 cl.D1 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.01% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The mammalian Phospholipase Cg (PLCg) family has two closely related proteins, PLCg1 and PLCg2. The PLCg isozymes have the core structure domains and a unique array of domains containing an additional PH domain, two SH2 domains and one SH3 domain. In response to extracellular stimuli, such as hormones and growth factors, receptor tyrosine kinases (RTKs) phosphorylate and activate PLCg. Activated PLCg catalyzes hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to produce the metabolic second messenger molecules inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG).
