          PPARGC1A Antibody (PA5-72948) in ICC/IF

          Immunocytochemistry analysis of PPARGC1A in HeLa cells. Samples were incubated with PPARGC1A polyclonal antibody (Product # PA5-72948) using a dilution of 1:200 dilution overnight at 4°C followed by anti-rabbit DyLight 488 (Green) at a dilution of 1:500. Cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100.           In IHC-P, staining is very strong in the nucleus with some cytoplasmic staining. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended.

          Target Information

          PGC1 alpha is a transcriptional coactivator that regulates the genes involved in energy metabolism. It interacts with PPARgamma, which permits it's interaction with multiple transcription factors. PGC-1 Alpha can interact with, and regulate the activities of, cAMP response element binding protein (CREB) and nuclear respiratory factors (NRFs). It provides a direct link between external physiological stimuli and the regulation of mitochondrial biogenesis, and is a major factor that regulates muscle fiber type determination. It may be also involved in controlling blood pressure, regulating cellular cholesterol homoeostasis, and the development of obesity.
