The TSCOT gene, known as the Thymic Stromal Co-transporter, plays a crucial role in the gene expression regulation within thymic epithelial cells, essential for T cell development. It encodes a protein that bears weak homology to bacterial 12 transmembrane co-transporters. Research indicates that TSCOT is predominantly expressed in thymic tissues, although low levels of expression are detectable in other epithelial tissues such as skin and lungs. The gene has been co-analyzed with Foxn1 and Hoxa3, highlighting its significant role in organogenesis. Studies utilizing competitive reverse transcription-polymerase chain reaction (RT-PCR) have mapped TSCOT to specific genomic regions, verifying its expression in a tissue and cell-type specific manner. The introduction of a neoantigen into TSCOT-expressing cells demonstrates its capability to mediate antigen-specific T cell tolerance, offering potential applications in immunological therapies.
