WUS encodes a plant-specific WUSCHEL-related homeobox transcription factor that localizes to the shoot apical meristem organizing center (niche) beneath the stem cells, and its protein can move from the organizing center into the adjacent central zone to execute non-cell-autonomous control of stem cell behavior. The gene produces a small, intron-containing transcript encoding an approximately 291-292 amino acid nuclear protein with a conserved N-terminal homeodomain responsible for sequence-specific DNA binding, plus a characteristic C-terminal WUS box required for its regulatory activities. Functionally, WUS is essential for establishing and maintaining the shoot stem cell niche: it promotes stem cell fate in overlying cells while simultaneously driving a homeostatic negative-feedback circuit by activating CLAVATA3 expression in neighboring stem cells, which in turn restricts WUS expression to prevent stem cell overproliferation; at the transcriptional level WUS also directly represses differentiation programs in meristematic progeny, thereby preventing premature organogenic differentiation and sustaining long-term meristem function.
