The Human Protein S (PROS) ELISA quantitates PROS in plasma
The Human Protein S solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the binding of the second (detector) antibody to the target on a different epitope from the capture antibody. An antibody conjugated with enzyme binds the formed sandwich. After incubation and washing steps to rid the microplate of unbound substances, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

Protein S is a vitamin K-dependent, potent neutral anticoagulant plasma glycoprotein. Protein S has a molecular weight of approximately 70 kDa and consists of a single polypeptide chain. The concentration of protein S in human plasma is 25 µg/mL, with a half-life of approximately two days. About 40% of Protein S in plasma is in a free form, whereas 60% is complexed with C4b-binding Protein (C4BP). Only the free Protein S functions as a cofactor to antigen presenting cells (APC) in the APC-dependent degradation of Factor Va and Factor VIIIa.
