CRHR1 Antibody in Immunocytochemistry (ICC/IF) — HEK 293 cells stably transfected with Corticotropin-Releasing Hormone Receptor 1 (CRHR1) were grown on poly-L-lysine-coated coverslips overnight. After 24 h, cells were fixed with 4% paraformaldehyde and 0.2% picric acid in phosphate buffer (pH 6.9) for 30 min at room temperature and washed several times with phosphate buffer. Specimens were permeabilized with ice-cold methanol and then incubated with Anti-CRHR1 Recombinant Rabbit Polyclonal Antibody (Product # 711642 at a concentration of 1 µg/mL). Cells were then incubated with Alexa488-conjugated secondary antibody for 2 h at room temperature, mounted and examined using a laser scanning confocal microscope. Note, strong staining of transfected cells.

CRHR1 Antibody in Western Blot (WB) — HEK 293 cells stably transfected with Corticotropin-Releasing Hormone Receptor 1 (CRHR1) were lysed in detergent buffer and glycoproteins were enriched using wheat germ lectin agarose beads. Receptors were eluted from the beads using SDS sample buffer for 20 min at 45 °C. Extracts were separated on 7.5% SDS-polyacrylamide gels and blotted onto PVDF membranes. Membranes were then incubated with Anti-CRHR1 Recombinant Rabbit Polyclonal Antibody (Product # 711642 at a concentration of 1 µg/mL). A broad band from 60-90 kDa corresponding to CRHR1 was observed.

Corticotropin-releasing factor receptor 1 (CRFR1/CRHR1), a Corticotropin-Releasing Factor Receptor, is involved in mediating the stress response and anxiety-related behaviors. Mutations of the receptor reduce anxiety-related behaviors and may be involved in the pathogenesis of Cushing disease. Multiple isoforms of CRFR1 are produced by alternative splicing. CRFR1 has been reported to be expressed in the pituitary, adrenal gland, brain, colon, ovary, placenta, testis, white blood cells, and skin. ESTs have been isolated from human brain, eye, colon, and embryo libraries.
