MDC1 Antibody in Western Blot (WB) — Western blot was performed using Anti-MDC1 Polyclonal Antibody (Product # PA1-41131) and a 250kDakDa band corresponding to Mediator of DNA damage checkpoint protein 1 was observed across the cell lines tested. Nuclear enriched extracts (30 µg lysate) of HeLa (Lane 1), A-431 (Lane 2), HCT 116 (Lane 3), Mouse Testis (Lane 4) and Rat Testis (Lane 5) were electrophoresed using NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Reagent Kit (Product # WP20005).

MDC1 participates in the early response to DNA damage. It is thought to be a member of the BRCT (BRCA1 C-terminus) super family of nuclear proteins. It contains an N-terminus forkhead-associated (FHA) motif, two C-terminus BRCT motifs and 13 internal repetitions of a 41 amino acid sequence.
