Phospho-CDK6 (Thr177) Antibody in Immunocytochemistry (ICC/IF) — For immunofluorescence analysis, T98G cells were fixed and permeabilized for detection of endogenous CDK6 (pT177) using Anti- CDK6 (pT177) Recombinant Rabbit Polyclonal Antibody (Product # 711588, 2 µg/mL) and labeled with Goat anti-Rabbit IgG (H+L) Superclonal Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034, 1:2000). Nuclei (blue) were stained using SlowFade® Gold Antifade Mountant with DAPI (Product # S36938), and Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300) was used for cytoskeletal F-actin (red) staining. Detection and localization of CDK6 pT177 (green) in the nucleus can be clearly observed in cells treated with forskolin (5 uM, 16 h) as compared to untreated cells. Antibody specificity was demonstrated by competition with the CDK6 pT177 peptide, which results in loss of signal. No competition was observed with the non-phospho peptide. The images were captured at 60X magnification.

Cyclin-dependent kinase 6 (p40cdk6 also known as PLSTIRE) belongs to a family of proteins designated as cdks. They are critical regulators of cell cycle progression. The prototype member of this family, p34cdc2, and a related protein cdk2, function late in the cycle while cdk4 and cdk6 are critically involved in G1 to S progression. CDK6 is essential for cell proliferation within the dentate gyrus of the hippocampus and the cuventricular zone of the lateral ventricles. Mutations in the gene results in microcephaly 12, primary, autosomal recessive (MCPH12).
