Essential 8™ 培养基是一种无异种成分、无饲养层的培养基,专为人多能干细胞 (PSC) 的生长和扩增而配制。Essential 8™ 培养基最初由 James Thomson 实验室的 Guokai Chen 等人所开发(以“E8”命名发表)并经 Cellular Dynamics International 所验证,目前已经得到了广泛的测试并证明可维持多种 iPSC 细胞系的多能性。此外,Essential 8™ 培养基已经被用来提高 iPSC 的产量,而且有证据表明其支持 iPSC 生长 >50 代而没有任何核型异常的迹象,同时还保持了 iPSC 分化为全部三系生殖谱系的能力。

可以。在mTeSR培养基+BD Matrigel基底膜基质条件下培养并冻存的PSCs细胞可复苏至Gibco Essential 8培养基中并种植于VTN-N基质上。某些细胞系复苏至冻存前的生长培养基及基质中可能会效果更好。在随后的下一次传代过程中,就可使用EDTA将这些细胞传代至Gibco Essential 8培养基+VTN-N的组合中了。
