Applied Biosystems™ TaqMan™ Advanced miRNA cDNA 合成试剂盒使用通用型逆转录 (RT) 化学试剂盒制备 cDNA 模板、以便与 TaqMan™ Advanced miRNA 测定一起用于检测和定量生物样本中成熟人 miRNA。

TaqMan Advanced miRNA cDNA 合成试剂盒使用的3'个 poly-A 拖尾和适配器序列的5'个连接用于逆转录前延伸样本中存在于末端的成熟 miRNA。通用 RT 引物识别存在于成熟 miRNA 的 5'和3'延伸末端上的通用序列。样本中所有成熟 miRNA 都被逆转录成 cDNA。

为了改善低表达 miRNA 靶标的检测,然后使用通用 miR-Amp 引物和 miR-Amp 预混液扩增 cDNA ,以均匀增加各个靶标 cDNA 的量,并保持相对差异表达水平。与传统预扩增不同,通用 miR-Amp 引物可识别添加至所有成熟 miRNA 的5'和3'末端的通用序列,确保不产生扩增偏倚。
