GSDMD (Cleaved Gly276) refers to the activated form of gasdermin D generated following proteolytic cleavage at glycine 276, a key step in the execution of pyroptosis. Gasdermin D is encoded by the GSDMD gene and is synthesized as a full-length protein composed of an N-terminal pore-forming domain and a C-terminal autoinhibitory domain. Upon activation of inflammatory caspases-most notably caspase-1 in canonical inflammasome pathways and caspase-4/5 (human) or caspase-11 (mouse) in noncanonical pathways-GSDMD is cleaved at a specific linker region near Gly276. This cleavage releases the N-terminal fragment, which oligomerizes and inserts into the plasma membrane to form pores, leading to cell swelling, membrane rupture, and the release of pro-inflammatory cytokines such as IL-1β and IL-18. Detection of cleaved GSDMD (Gly276) is widely used as a molecular marker of inflammasome activation and pyroptotic cell death in inflammation and innate immunity research.
