Immunohistochemistry analysis of paraffin-embedded Phospho-MLK3 (Thr277, Ser281) in human pancreas tissue sections. Antigen retrieval was performed using citrate buffer. Samples were blocked with blocking buffer (1.5 hr, 22°C), incubated with Phospho-MLK3 (Thr277, Ser281) polyclonal antibody (Product # PA5-105817) using a dilution of 1:200 (1.5 hr, 22°C), followed by HRP conjugated goat anti-rabbit.

Antibody detects endogenous levels of MLK3 only when phosphorylated at Thr277+Ser281.
Target Information
MAP3K11 (MLK3) is a serine/threonine kinase that activates both the p38 MAP kinase and c-Jun kinase pathways. This kinase contains a SH3 domain and a leucine zipper-basic motif. This kinase preferentially activates MAPK8/JNK kinase, and functions as a positive regulator of JNK signaling pathway. This kinase can directly phosphorylate, and activates IkappaB kinase alpha and beta, and is found to be involved in the transcription activity of NF-kappaB mediated by Rho family GTPases and CDC42.
