The eBioscience BrdU Staining Kit for Flow Cytometry contains the necessary reagents and buffers for identifying and examining proliferating cells of mammalian species by flow cytometric analysis. Cycling cells are incubated with 5-bromo-2’-deoxyuridine (BrdU), a synthetic analog of thymidine which incorporates into newly synthesized genomic DNA during the S-phase of mitosis. Following DNA denaturation, the cells are stained for BrdU incorporation along with any other cell surface and/or intracellular targets of interest.

This kit is optimized to achieve brighter staining compared to traditional protocols.

Yes, primary antibodies against additional intracellular targets can be added alongside the Anti-BrdU antibody. However, it is critical that you empirically determine whether these additional antibodies are compatible with this staining protocol.

Is the BrdU in the eBioscience BrdU staining kit supplied as a sterile formulation for in vivo use?

Yes, the BrdU is supplied as a sterile liquid. The vial should be opened under sterile conditions if it will be used in vivo.

Can I keep the diluted (1 mM) BrdU in the eBioscience BrdU staining kit at 4 degrees C? Can I refreeze it after use?

The stock (32.5 mM) BrdU can be frozen and thawed up to three times. Furthermore, the stock BrdU can be kept at 4 degrees C for up to one week. However, the diluted (1 mM) BrdU should be used immediately.

Have the BrdU Staining kits been tested on proliferating cells of different species?

We have used this kit to detect proliferating human and mouse cells. Other species have yet to be tested but feline, non-human primate and canine should also work.

What are the stopping points during flow cytometric analysis when using a BrdU staining kit?

Samples can be stored at 4 degrees C for up to 16 hours (i.e., overnight) after fixation with BrdU Staining Buffer (with or without washing in Flow Cytometry Staining Buffer) with no adverse effect on performance.
