The Ion Torrent Genexus GX5 Starter Pack for Ion AmpliSeq panels provides one set of all core consumables required to run the Ion Torrent Genexus Integrated Sequencer. This starter pack supports flexible, fully automated preparation and normalization of up to 32 Ion AmpliSeq libraries, as well as sequencing for up to eight chip lanes (two full chips). The Ion Torrent GX5 Chip is used in conjunction with this starter pack and must be ordered separately.

As part of the Genexus Integrated Sequencer GX5 workflow, the Genexus GX5 Starter Pack-AS:
• Automates library preparation, templating, and sequencing for 200–400 base-read Ion AmpliSeq libraries
• Supports intake variability of clinical research samples in a cost-effective manner
• Maximizes flexibility with the ability to sequence one to four chip lanes per run
• Enables parallel processing of up to four compatible assays in a single run
• Delivers fast and simple instrument set-up, with less than five minutes of hands-on time

The Ion Torrent Genexus Integrated Sequencer automates library preparation, sequencing, analysis, and reporting in a hands-off, walkaway workflow to deliver results in a single day. The innovative, multilane sequencing chip is designed to enable cost-effective processing of 1 to 32 single pool libraries.

For Research Use Only. Not for use in diagnostic procedures.

Frequently asked questions (FAQs)

What are the correct shipping conditions for the Genexus Cartridges that are a part of the Genexus Sequencing Kit (Cat. No. A40271) and Genexus GX5 Starter Packs (Cat. Nos. A40279, A40280)?

The Genexus Cartridge should be shipped on supergels at -15 to -50 degrees C, in a polystyrene box. If the cartridges are shipped with dry ice or arrive thawed, they should not be used, and replacement would need to be issued.

How many samples of Oncomine Precision Assay GX can I run on each lane of the GX5 Chip?

The capacity of the GX5 Chip varies depending on the sample type:
- For plasma cfTNA samples, the workflow is configured to accommodate 1 sample per GX5 Chip lane (or 4 samples on a single GX5 Chip).
- For FFPE DNA+RNA samples, the workflow is configured to accommodate 4 samples per GX5 Chip lane.
- For FFPE DNA-only or RNA-only samples, the workflow is configured to accommodate 8 samples per GX5 Chip lane.

How many sequencing reactions can I perform with Oncomine Precision Assay GX (Cat. No. A44350)?

The maximum number of samples that can be analyzed per kit depends upon the type of analysis that you want to perform. Each kit provides the targeted pancancer panel and library reagents sufficient to perform up to 32 sequencing reactions on the Genexus Integrated Sequencer.

- Simultaneous DNA- and RNA-based variant analysis for up to 16 FFPE tissue samples (one DNA reaction and one RNA reaction per sample)
- DNA-only or RNA-only variant analysis for up to 32 FFPE tissue samples
- Cell-free TNA (cfTNA) variant analysis (combined DNA and RNA) for up to 32 plasma samples

How much of each sample do I need for the Oncomine Precision Assay GX?

For solid tumor samples, the optimal input amount is 10 ng with validated input in the range of 5-15 ng. For liquid biopsy samples, the optimal input amount is 20 ng with validated input in the range of 5-40 ng.

For optimal results, load 20 µL of input with a concentration of ≥0.67 ng/µL for FFPE DNA and FFPE RNA samples and 20 µL of input with a concentration of ≥1.33 ng/µL for cfTNA samples. If the concentration of the sample is lower than optimal, use as much input as possible for each reaction.

What controls should I use with Oncomine Precision Assay GX?

Here are the various controls we recommend depending upon the starting material:

FFPE DNA samples
- Structural Multiplex Reference Standard (FFPE), Cat. No. HD789 (Horizon™)
- EGFR Gene-Specific Multiplex Reference Standard (FFPE) 5% AF, Cat. No. HD300 (Horizon™)
- KRAS Gene-Specific Multiplex Reference Standard (FFPE), Cat. No. HD301 (Horizon™)
- Seraseq® Lung & Brain CNV Mix, + 3 copies, Cat. No. 0710-0414 (SeraCare®)
