Detect low density lipoprotein (LDL) uptake, endosomal maturation, and trafficking in cells with pHrodo Green-LDL and pHrodo Red-LDL conjugates. These LDL conjugates contain pH-sensitive pHrodo dyes that are weakly fluorescent at neutral pH, yet fluoresce brightly upon exposure to acidic environments, such as those found in endosomes. The pHrodo Green-LDL and Red-LDL conjugates can be used in fluorescence microscopy, flow cytometry, high-throughput screening, and high content analysis experiments.
Human low density lipoprotein (LDL) is one of the key lipid:protein complexes in blood and is a crucial component of metabolism responsible for the transport of lipids throughout the body. LDL from human plasma, when complexed with pHrodo Green dye (pHrodo Green-LDL) or pHrodo Red dye (pHrodo Red-LDL), is a valuable tool in studies of LDL uptake through endocytosis and trafficking of LDL throughout the cell. pHrodo Green-LDL and pHrodo Red-LDL are dimly fluorescent at neutral pH outside of cells but fluoresce brightly after endocytosis. This feature allows for a no-wash protocol when using pHrodo Green-LDL or pHrodo Red-LDL, enabling kinetic measurement of LDL uptake, endosomal maturation, and trafficking in cells. These fluorescently labeled LDL conjugates have been designed for use with fluorescence microscopy, flow cytometry, cell sorting, high-throughput analysis, and high content analysis.

Features of pHrodo Green-LDL:
• Detectable with GFP/FITC filter set (absorption peak: ∼509 nm, emission peak: ∼533 nm)
• Mutiplexable with a wide variety of blue, red, and far-red dyes reporters such as Mitosox Red, CellEvent Caspase 3/7 Red, NucBlue, RFPs, and Mitotracker Deep Red, among many others.

Features of pHrodo Red-LDL:
• Detectable with RFP/TRITC filter set (absorption peak: ∼560 nm, emission peak: ∼585 nm)
• Mutiplexable with a wide variety of blue, green, and far red dyes and reporters such as GFP, Fuo-4, calcein, NucBlue, CellEvent Caspase 3/7 green, Mitosox Green, and Mitotracker Deep Red, among many others.

Both pHrodo Green-LDL and pHrodo Red-LDL can be used in a no-wash protocol, allowing dynamic measurement of LDL uptake and cell trafficking.

This product does not contain acetylated or oxidized LDL.

Why is fixation not recommended for cells stained with pHrodo Red-LDL?

pHrodo Red and Green dyes are pH-sensitive dyes. They should only be used on live cells to detect pH of cellular compartments. Upon fixation of the cells, the cell membrane is compromised and you are only measuring the pH of the buffer the samples are held in.
