To manufacture the standards, GMO flour is blended with non-GMO flour to obtain specific percentage GMO content, in the range of 0% to 5%. In addition, these standards can serve as a template preparation control since the template for the assay is prepared directly from the soy or maize flour.
Are GMO maize or GMO soy reference standards included in the TaqMan GMO Maize 35S (P/N 4327693) and the TaqMan GMO Soy 35S (P/N 4327692) kits?
No, reference standards are not provided in these kits. The standards are manufactured by the Institute of Reference Materials and Measurements (IRMM), and are available through distributors such as Fluka and Resource Technology Corporation.
How does one calculate the amount of genetically modified organisms (GMO) present in a food sample?
The TaqMan GMO Detection Kits are multiplex assays that detect and quantify a GMO-specific target and a plant-specific target. The amount of GMO soy or GMO maize is calculated as a percent of the total maize or soy in any given food. The endogenous reference target provides a means for determining the total amount of maize or soy in the sample. Including the appropriate GMO standard permits accurate determination for the percent of GMO content in each sample. The absence of a signal from the endogenous target, indicating the presence of PCR inhibitors in the sample, signifies a false negative result. What target sequences do the TaqMan GMO (genetically modified organisms) kits detect?
In both of the kits: the TaqMan GMO Maize 35S (P/N 4327693) and the TaqMan GMO Soy 35S (P/N 4327692) Detection Kits, there is a GMO-specific target and a specific endogenous reference (ER) target. The GMO-specific target for both kits is the Cauliflower Mosaic Virus (CMV) 35S promoter. The ER target is a maize- or soy-specific locus. The ER target acts both as an internal positive control and as a means for determining the total amount of maize or soy in the sample.
