LIVE/DEAD 活力/细胞毒性测定试剂盒(绿色/深红色)是一种双色荧光细胞活力测定试剂盒,可同时鉴别活细胞和死细胞。活细胞用钙黄绿素染料标记,并发出绿色荧光,而死细胞用 SYTOX 深红色染料标记,并发出红色荧光。通过可兼容的滤光片组,这些染料可与其他橙色/红色荧光基团联用。

• 通过双色荧光测定细胞活力
• 使用 GFP/FITC 滤光片组检测活细胞,使用深红色/CY5 滤光片组检测死细胞
• 可与橙色/红色荧光基团联用,例如 RFP、mCherry、Mitotracker 红色/橙色、Lysotracker 红色、TMRM 等

可通过几乎不发出荧光且可透过细胞的钙黄绿素 AM 和高强度荧光钙黄绿素的酶催化转化,确定细胞内普遍存在的酯酶活性,从而区分活细胞。钙黄绿素在活细胞内保存完好,且会在活细胞中产生强烈且均匀的绿色荧光(激发/发射波长为 ∼495/∼515 nm)。使用传统 FITC/GFP 滤光片组可检出钙黄绿素。SYTOX Deep Red 染料可进入细胞膜受损的细胞,与 DNA 结合后荧光增强数倍。死亡或质膜受损的细胞核可发出明亮的荧光,使用 CY5/深红色滤光片组(激发/发射波长最大值:660/682 nm)。

该试剂盒适合与荧光显微镜或高内涵分析 (HCA) 荧光多壁板扫描仪配合使用,且可轻松调整,以与其他荧光检测系统配合使用。测定原理具有通用性,适用于大多数真核细胞类型,包括贴壁细胞和非贴壁细胞。

We have not validated the use of LIVE/DEAD Viability/Cytotoxicity Assay Kit (Green/Deep Red)(Cat. No. L32250) on bacteria, fungi or protozoa. In general, calcein, AM does not work well with bacteria due to their low intracellular esterase activity. Some protozoa may readily engulf cell impermeant stains and result in intracellular staining of live cells.
