细胞成像应用
Hoechst 33342(2'-[4-乙氧基苯基]-5-[4-甲基-1-哌嗪基]-2,5'-二-1H-苯并咪唑)的光谱特性,包括较大的斯托克斯位移,使其非常适合与绿色(Alexa Fluor™ 488、FITC、GFP)和红色(Alexa Fluor™ 594、Texas Red™、罗丹明、mCherry、mKate-2)荧光基团一起用于多色实验。由于 Hoechst 33342 对 DNA 的亲和力高,因此也常用于细胞计数、细胞周期和细胞复制研究中识别凋亡细胞的浓缩细胞核,与 Click-iT™ EdU 或 BrdU 染色联合用于细胞周期研究,作为高内涵成像分析中的细胞核分割工具,还可根据细胞 DNA 含量分选细胞。

使用建议
NucBlue™ 活细胞染色剂可直接添加到完全培养基或缓冲溶液中的细胞中。
• 在大多数情况下,2滴/mL 以及15至30分钟的孵育时间将会产生明亮的细胞核染色;然而,可能需要针对一些细胞类型、条件和应用进行优化。在这种情况下,只需增加或减少滴数,直至获得最佳染色强度即可。在大多数情况下,如果在成像前不洗涤细胞,染色强度随时间的增加而增加。
•与 DNA 结合后,NucBlue™ 活细胞染色剂可被 360 nm 的紫外光激发,最大发射波长为 460 nm。可通过蓝/青色滤光片进行检测,如 DAPI 滤光片、蓝色 GFP 滤光片或 Semrock BrightLine™ Alexa Fluor™ 350 染料滤光片套件。

Can I use the ReadyProbes reagents for flow cytometry?

This is not recommended. The ReadyProbes reagents were developed for imaging applications whereas the Ready Flow reagents were optimized for flow cytometry.
