ALKBH4 Antibody (PA5-100872) in ICC/IF
Immunocytochemistry analysis of ALKBH4 in A549 cells. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25°C. Samples were incubated with ALKBH4 polyclonal antibody (Product # PA5-100872) and mouse anti-beta tubulin antibody for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) (Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) (Green) were used as the secondary antibodies. The nuclear counter stain is DAPI (blue).

Target Information
Dioxygenase that mediates demethylation of actin monomethylated at 'Lys-84' (K84me1), thereby acting as a regulator of actomyosin-processes (PubMed:23673617). Demethylation of actin K84me1 is required for maintaining actomyosin dynamics supporting normal cleavage furrow ingression during cytokinesis and cell migration (PubMed:23673617). In addition to proteins, also demethylates DNA: specifically demethylates DNA methylated on the 6th position of adenine (N(6)-methyladenosine) DNA, thereby regulating Polycomb silencing. [UniProt]
