          Target Information

          Protein Kinase C iota (PKC iota), also known as PRKCI, is an atypical isoform of Protein Kinase C that plays significant roles in cellular signaling. The PKC iota gene is positioned on the human chromosome 3, specifically at 3q26. Its structure is characterized by an absence of the calcium and diacylglycerol regulatory domains that are typically found in conventional PKC isoforms, allowing it to function independently of these signals. PKC iota has been identified as a bona fide oncogene, implicated in various cancers, including lung and ovarian cancers. It is involved in biological processes such as cell differentiation, proliferation, and survival by influencing pathways like NF-?B activation. PKC iota interacts with TRAF6 and IRAK to activate I?B kinase-β, prompting NF-?B to translocate to the nucleus and promote gene expression related to cell survival and growth. Molecular cloning studies have detailed its structure, which is pre-organized before substrate binding, differing considerably from other kinase family members like PKA. PKC iota's critical role in cancer progression and its potential as a prognostic marker make it a key target for therapeutic research, particularly in oncogenic pathways linked to K-Ras mutations.
