Immunocytochemistry analysis of 53BP1 in Neuro2A cells. Samples were incubated with 53BP1 polyclonal antibody (Product # PA5-22754) using a dilution of 2 µg/mL overnight at 4°C followed by anti-rabbit Dylight 488 (Green) at a dilution of 1:500. Cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton-X100. 53BP1 (P53-binding protein 1) plays a critical role in tumor suppression and is a putative substrate of ATM kinase. Upon DNA damage, 53BP1 is phosphorylated and localizes to the presumptive sites of damage, specifically, double-strand breaks. 53BP1 may have a role in checkpoint signaling during mitosis, enhances TP53-mediated transcriptional activation, and participates in DNA repair by maintaining genomic stability.
