          SOX9 Antibody (740051MP555) in ICC/IF

          Immunofluorescent analysis of SOX9 was performed using SOX9 Recombinant Mouse Monoclonal Antibody (GMPR9), Alexa Fluor™ Plus 555 (Product # 740051MP555, 1:200) on 70-80% confluent PC-3 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton X-100 for 15 minutes and blocked with 2% BSA for 1 hour at room temperature.           Alexa Fluor™ Plus recombinant antibodies are conjugated using new, proprietary dye chemistry so you can generate stunning data. Alexa Fluor™ Plus antibodies represent an advancement in fluorescent conjugate technology. Alexa Fluor™ Plus antibodies provide brighter signal compared to leading Alexa Fluor™ antibodies, providing you with better signal-to-noise for your critical experiments. These antibodies show better specificity and lot-to-lot consistency as these are recombinant antibodies, generated by cloning specific genes for the desired antibodies into an expression vector and expressed in vitro.

          Using conjugate solutions: Centrifuge the protein conjugate solution briefly in a microcentrifuge before use; add only the supernatant to the experiment. This step will help eliminate any protein aggregates that may have formed during storage, thereby reducing nonspecific background staining.

          Applications Tested: This GMPR9 antibody has been tested by immunocytochemistry and flow cytometric analysis of PC-3 cells. This may be used for immunocytochemistry at 5 µg/mL and for flow cytometry at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

          Target Information

          SOX9 has a role in sex determination and differentiation of Sertoli cells. It is involved in chondrogenesis and regulates the expression of other genes involved in chondrogenesis by acting as a transcription factor for these genes. Translocation of this gene can cause campomelic dysplasia. SOX9 is involved in the formation of testes from the indifferent fetal gonads. It is a major molecular component of the neuron-glia switch in developing spinal cord. During normal development, SOX9 allows the prostate epithelium to outgrow into the mesenchyme and then provides basal cell support for development and maintenance of the luminal epithelium. These functions of SOX9 are subverted in prostate cancer to support tumor growth and invasion. SOX9 may direct the formation of neural crest precursors and the development of a range of neural crest derivative. It has a transcriptional regulation in melanin production in cells. Northern blot analysis shows its expression on adult testis, adult heart, and fetal brain.
