ACD1 encodes a chloroplast-associated enzyme of the chlorophyll catabolic pathway that localizes to plastids (consistent with a chloroplast transit peptide) and functions during leaf senescence and stress-related chlorophyll breakdown. The ACD1 protein is a Rieske-type oxygenase/monooxygenase of the pheophorbide a oxygenase (PaO) step, containing a Rieske 2Fe-2S cluster-binding module and a non-heme iron catalytic center typical of aromatic-ring-hydroxylating dioxygenase-like enzymes, enabling oxygenolytic opening of the pheophorbide a tetrapyrrole macrocycle. Biologically, ACD1 activity is required to convert pheophorbide a into downstream chlorophyll catabolites; when ACD1/PaO activity is reduced (e.g., by antisense suppression), pheophorbide a accumulates and triggers cellular damage and accelerated cell death, including photooxidative injury upon re-illumination after dark-induced senescence, demonstrating that controlled PaO function is essential both for efficient chlorophyll detoxification during senescence and for limiting pro-death signaling/toxicity from chlorophyll catabolic intermediates.
