Western blot analysis was performed on whole cell extracts (30 µg lysate) of MCF-7 (Lane 1), NTERA-2 cl.D1 (Lane 2), HEL 92.1.7 (Lane 3), K-562 (Lane 4), MOLT4 (Lane 5), A-431 (Lane 6), THP-1 (Lane 7), MDA-MB-231 (Lane 8) and T-47D (Lane 9). The blots were probed with Anti-AGO1 Recombinant Rabbit Superclonal™ Antibody (Product # 711755, 1-2 µg/mL). A 97 kDa band corresponding to AGO1 was observed across most of the cell lines tested. AGO1 is a member of the Argonaute family of proteins which play a role in RNA interference. The protein is highly basic, and contains a PAZ domain and a PIWI domain. It may interact with dicer1 and play a role in short-interfering-RNA-mediated gene silencing. AGO1 binds to miRNAs or siRNAs and represses the translation of mRNAs.
