Argonaute 1-PAZ encodes the PAZ (Piwi/Argonaute/Zwille) RNA-binding module found within Argonaute-family effector proteins, which localize predominantly to the cytoplasm as core components of the RNA-induced silencing complex (RISC) and can also participate in nuclear RNA-directed silencing. Structurally, the PAZ region is a compact globular fold built around a left-handed six-stranded beta-barrel capped by two alpha-helices and augmented by a distinctive beta-hairpin/short alpha-helix appendage that helps form an RNA-binding pocket. Functionally, the PAZ domain binds the 3-prime end of small RNAs (miRNAs/siRNAs), helping position guide RNAs within Argonaute and supporting key steps in RISC maturation and activity; genetic and biochemical studies indicate that PAZ integrity is particularly important for slicing-independent RISC activation, where it promotes passenger-strand ejection/unwinding and formation of a functional, single-stranded guide-containing complex.
