Argonaute 10 (AGO10) encodes a cytoplasmic/nucleocytoplasmic Argonaute-family RNA-binding protein that functions within small RNA-directed silencing pathways and is particularly important in meristematic and organ polarity contexts. AGO10 is a multidomain, bilobed protein built around the conserved Argonaute core architecture comprising an N-terminal region followed by structured linker segments that position the PAZ, MID, and PIWI domains; the PAZ domain forms a pocket that recognizes the 3' end of small RNAs, the MID domain helps anchor the 5' end, and the PIWI domain adopts an RNase H-like fold associated with endonucleolytic "slicer" potential in Argonautes. Biologically, Arabidopsis AGO10 is a key regulator of shoot apical meristem maintenance and leaf adaxial-abaxial patterning by preferentially binding and sequestering miR165/166, thereby limiting their availability to AGO1-containing effector complexes and relieving repression of class III HD-ZIP transcripts; genetic and biochemical data support a primary "decoy/sequestration" role for AGO10's small-RNA binding activity in development, with catalytic activity not being the essential requirement for this function.
