AHB1 encodes a class 1 non-symbiotic plant hemoglobin (globin) that is primarily localized in the cytosol and accumulates in specific aerial tissues (including leaf hydathodes and developing floral organs) as well as being inducible by low-oxygen conditions. The protein adopts the conserved 3-on-3 alpha-helical globin fold and binds a b-type heme cofactor; it is characterized by predominantly bis-histidyl (hexacoordinate) heme ligation yet can reversibly bind diatomic ligands, conferring extremely high oxygen affinity that argues against a role as an oxygen transporter. Functionally, AHB1 acts as a key modulator of nitric oxide (NO) homeostasis: genetic and biochemical studies show it scavenges NO (forming S-nitrosohemoglobin/iron-nitrosyl species), thereby reducing NO emission during hypoxic stress and limiting NO accumulation in tissues where it is expressed, with downstream effects on growth and stress physiology; in addition, deoxygenated AHB1 can catalyze nitrite reduction to NO in vitro, supporting a role in integrating oxygen status with reactive nitrogen metabolism.
