AHK2 encodes a membrane-bound hybrid sensor histidine kinase cytokinin receptor in Arabidopsis thaliana that is found predominantly on endomembranes, with strongest evidence for localization to the endoplasmic reticulum. The protein is a single-pass/multi-pass transmembrane receptor-like kinase with an N-terminal extracytosolic sensory region containing a ligand-binding CHASE domain, followed by transmembrane segments that anchor it in the membrane and a C-terminal cytosolic signaling module comprising the conserved histidine kinase core (including the phosphoacceptor and ATPase subdomains) fused to a receiver (REC) domain, enabling two-component style phosphotransfer. Functionally, AHK2 binds bioactive cytokinins (with high affinity reported for isopentenyladenine and trans-zeatin) to promote receptor autophosphorylation and initiation of a multistep His-to-Asp phosphorelay through histidine phosphotransfer proteins to response regulators, thereby regulating cytokinin-dependent transcriptional programs that influence meristematic activity, organ growth, root and shoot development, and physiological stress responses, acting partially redundantly but with distinct spatial activity compared with the other cytokinin receptors.
