Aristolochic acids are a group of structurally related nitrophenanthrene carboxylic acid derivatives found naturally in the Aristolochiaceae plant family, traditionally used in herbal medicines. The distinctive structure includes a phenanthrene core with a nitro group, which contributes to their highly toxic and carcinogenic properties. Aristolochic acids are known to form DNA adducts, leading to mutations and promoting tumorigenesis, which is linked to urothelial cancer and nephropathy known as Aristolochic Acid Nephropathy (AAN). Due to their genotoxic effects, the use of plants containing these compounds has been heavily scrutinized, resulting in regulatory actions to limit exposure and ensure consumer safety. Despite their historical use for various ailments, research underscores the critical need to understand the mechanisms of toxicity and implement stringent controls in herbal product manufacturing to prevent adverse health outcomes.
