B7-H3 Antibody (PA5-47132) in WB
Western blot analysis of B7-H3 lysates of MEF mouse embryonic feeder cells, P19 mouse embryonal carcinoma cell line, NIH-3T3 mouse embryonic fibroblast cell line, C2C12 mouse myoblast cell line, and 3T3-L1 mouse embryonic fibroblast adipose-like cell line. PVDF membrane was probed with 1 µg/mL B7-H3 Polyclonal Antibody (Product # PA5-47132) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody. A specific band was detected for B7-H3 at approximately 4555 kDa (as indicated). This experiment was conducted under reducing conditions.

In direct ELISAs and Western blots, approximately 50% cross-reactivity with recombinant human (rh) B7-H3 is observed, and less than 1% cross-reactivity with recombinant mouse (rm) B7-H1, rmB7-H2 and rmB7-1 is observed.
Reconstitute at 0.2 mg/mL in sterile PBS.
Endoxin level is <0.10 EU per 1 µg of the antibody by the LAL method.
Target Information
CD276 is a member of the B7 family of co-stimulatory molecules also known as B7-H3. CD276 is a type I transmembrane protein that induces the proliferation of CD4+ and CD8+ T cells, enhances the generation of cytotoxic T cells and selectively stimulates the production of interferon gamma. Expression of CD276 can be induced on dendritic cells and monocytes by inflammatory cytokines, and is also widely expressed in peripheral tissues including the heart, kidney, testis and colon. In humans, CD276 exists as two isoforms which result from gene duplication and differential splicing. CD276 is reported to have therapeutic potential for the regulation of cell-mediated immune responses to cancer, particularly in conjunction with anti-angiogenic therapy.
