B7-H7 Antibody (61-6537-42) in Flow

HDLM-2 cells were stained with Mouse IgG1 kappa Isotype Control, PE-eFluor 610 (Product # 61-4714-82) (blue) or B7-H7 Monoclonal Antibody, PE-eFluor 610 (purple). Total viable cells, as determined by Fixable Viability Dye eFluor 450 (Product # 65-0863-18), were used for analysis.

B7-H7 Antibody — Surface staining of PBMC’s, HDLM-2, and HeLa cells. As expected based on known relative expression patterns, B7-H7 clone MA57YW stains HDLM-2 cells, but not HeLa cells or human PBMC’s. Details: Staining of human peripheral blood cells (top left) were co-stained with CD14 Monoclonal Antibody, (clone 61D3) and CD3 Monoclonal Antibody, (clone OKT3). PBMC’s were stained with Mouse IgG1 kappa Isotype Control (clone P3.6.2.8.1) (blue) or B7-H7 Monoclonal Antibody. Cells were gated on CD3 positive CD14 negative (purple top right), CD3 negative and CD14 positive (blue middle left), and CD3 negative (orange middle right). HeLa (bottom left histogram), or HDLM-2 (bottom right histogram) cells were stained with Mouse IgG1 kappa Isotype Control (blue) (clone P3.6.2.8.1) or B7-H7 Monoclonal Antibody, (purple). Description: This MA57YW monoclonal antibody recognizes human B7-H7 on live and paraformaldehyde-fixed cells.

Applications Reported: This MA57YW antibody has been reported for use in flow cytometric analysis. This MA57YW monoclonal antibody recognizes human B7-H7 on live and paraformaldehyde-fixed cells.

Applications Tested: This MA57YW antibody has been pre-diluted and tested by flow cytometric analysis the HDLM-2 cell line. This may be used at 5 µL (0.5 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.

PE-eFluor 610 can be excited with laser lines from 488-561 nm and emits at 607 nm. We recommend using a 610/20 band pass filter (equivalent to PE-Texas Red). Please make sure that your instrument is capable of detecting this fluorochrome.

Light sensitivity: This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light.

Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.

B7-H7 (also known as HHLA2) is a membrane protein with three Ig-like domains (IgV-IgC-IgV), whereas other members of the B7 family generally have only two Ig domains (IgV-IgC). B7-H7 is somewhat more closely related to B7-H3 and B7-H4 and shares 10% to 18% amino acid identity and 23% to 33% similarity to B7 family members. B7-H7 protein is widely expressed in epithelium cells of the gallbladder, breast, kidney, colon, small intestines and placenta. In the immune system, HHLA2 protein is not constitutively expressed on human monocytes/macrophages. B7-H7 is widely expressed in cancers, especially in triple negative breast cancer.
