The BRD9 gene encodes a protein that is a crucial subunit of the SWI/SNF chromatin remodeling complex known as ncBAF (non-canonical BAF complex). As a bromodomain-containing protein, BRD9 acts as an 'epigenetic reader,' selectively recognizing acetyl-lysine marks on histones, which is essential for regulating chromatin structure and gene expression through the recruitment of acetylated transcriptional regulators. It has emerged as a significant player in epigenetic modulation and has been implicated in various cancers, such as cervical and non-small cell lung cancer, due to its over-expression. BRD9 is situated on the 5p arm of chromosome 5 and is a potential target for anticancer therapies, notably in acute myeloid leukemia (AML). Recent studies highlight the development of selective BRD9 inhibitors, indicating its potential for therapeutic targeting. Its role in cancer is linked to its function in transcriptional regulation and its involvement in the mammalian SWI/SNF complex, which is linked to tumor suppression mechanisms.
