BRI1 encodes a plasma membrane, single-pass transmembrane leucine-rich repeat receptor-like serine/threonine kinase that functions as a principal cell-surface receptor for brassinosteroid hormones in plants. The protein comprises an N-terminal signal peptide and a large extracellular ectodomain built from a superhelix of ~25 leucine-rich repeats that includes a distinctive ~70-amino-acid "island" insertion; this island and adjacent leucine-rich repeats form the steroid-binding pocket for brassinolide. A short transmembrane helix anchors the receptor, followed by a cytosolic kinase domain whose activation depends on phosphorylation and receptor complex formation. Upon brassinosteroid binding, BRI1 promotes assembly with a SERK-family co-receptor (notably BAK1), enabling reciprocal trans-phosphorylation of the cytoplasmic kinase domains and initiation of downstream signaling that alters transcriptional programs controlling broad aspects of growth and development (including cell expansion and developmental patterning).
