BZR1 encodes a nucleus-localized, brassinosteroid-responsive transcription factor that functions as a central downstream effector of the plasma-membrane brassinosteroid receptor pathway, shuttling between cytoplasm and nucleus in a phosphorylation-dependent manner. The Arabidopsis BZR1 protein is ~336 aa and contains an N-terminal BES1_N/BZR1-type DNA-binding domain, multiple regulatory phosphorylation sites targeted by the GSK3-like kinase BIN2, and a PEST-rich region implicated in protein turnover and regulatory interactions. In low brassinosteroid conditions, BIN2 phosphorylation reduces BZR1 activity by diminishing DNA binding, promoting association with 14-3-3 proteins that favors cytoplasmic retention, and increasing proteasome-mediated degradation; upon pathway activation, BZR1 becomes dephosphorylated (notably by PP2A), accumulates in the nucleus, and directly modulates brassinosteroid-responsive gene expression programs that drive growth and developmental outputs.
