Bleomycin resistance protein is a small intracellular bleomycin-binding determinant found in several bacteria (including bleomycin-producing actinomycetes and mobile genetic elements), where it functions primarily by drug sequestration rather than enzymatic modification. Structural studies show a compact, predominantly alpha/beta protein of ~14 kDa (about 120-130 aa) that forms a stable homodimer via domain/arm exchange, creating a groove/crevice at the dimer interface that constitutes the principal bleomycin-binding site; in solution, the functional dimer can bind two bleomycin molecules, thereby preventing the antibiotic from engaging DNA and suppressing bleomycin-dependent DNA strand scission. In cellular contexts, bleomycin resistance mediated by this binding protein can be complemented by host DNA repair capacity, consistent with the idea that rapid intracellular neutralization of bleomycin reduces genotoxic burden and improves survival under exposure.
