YFP, Yellow Fluorescent Protein, is a bright variant of green fluorescent protein optimized for yellow emission, widely used as a noninvasive reporter in plant biology. It fluoresces without added cofactors, enabling live imaging of protein localization, organelle dynamics, and gene expression in tissues from roots to leaves. YFP fusions track protein trafficking and interactions, while targeted variants label nuclei, membranes, cytoskeleton, chloroplasts, and mitochondria. Its excitation and emission are compatible with common confocal and widefield microscopes and allow multiplexing with CFP, GFP, mCherry, and red chlorophyll autofluorescence. Plant optimized YFP versions offer improved folding, photostability, and pH tolerance, and can be expressed under constitutive or inducible promoters. YFP is also used in FRET assays to quantify protein interactions and signaling kinetics in vivo, making it a versatile tool for visualizing cellular processes and developmental patterns in plants.
