Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please, remember to spin tubes briefly prior to opening them to avoid any losses that might occur from lyophilized material adhering to the cap or sides of the tubes.

CALS12 encodes a large integral membrane callose synthase that localizes predominantly to the plasma membrane (and is also reported in Golgi-associated fractions), where it functions within a 1,3-beta-D-glucan synthase complex to polymerize callose for deposition at the cell wall-plasma membrane interface and at plasmodesmata. The protein is predicted to be multi-pass, with multiple C-terminal transmembrane helices anchoring a substantial cytosolic catalytic region characteristic of glucan synthase-like/glycosyltransferase enzymes. Biologically, CALS12-dependent callose accumulation contributes to regulated cell-to-cell connectivity by modulating plasmodesmatal permeability and reinforces local wall barriers during stress; genetic and physiological studies support key roles in inducible callose deposition during pathogen challenge (impacting penetration resistance and defense signaling outputs) and in adaptive responses such as phosphate deficiency, where CALS12 is required for callose deposition in root hairs that supports phosphate acquisition and growth under low-phosphate conditions.
