Caspase-1 p10 (cleaved Ala317) refers to the small, active subunit generated following proteolytic processing of pro-caspase-1, an inflammatory cysteine protease encoded by the CASP1 gene. Caspase-1 is synthesized as an inactive zymogen that becomes activated within multiprotein inflammasome complexes in response to pathogenic infection or cellular stress. Activation involves autocatalytic cleavage at specific aspartic acid residues, including near alanine 317, resulting in the formation of the p20 (large) and p10 (small) subunits that assemble into the catalytically active enzyme. The cleaved p10 fragment is therefore a marker of caspase-1 activation. Active caspase-1 mediates the maturation and secretion of pro-inflammatory cytokines such as interleukin-1β (IL-1β) and interleukin-18 (IL-18), and plays a central role in pyroptosis, a form of inflammatory programmed cell death. Detection of cleaved caspase-1 p10 (Ala317) is widely used as an indicator of inflammasome activation in immunology and inflammation research.
