Staining of thioglycolate-induced peritoneal exudate cells with 0.25 µg of Rat IgG2a K Isotype Control FITC (Product # 11-4321-42) (open histogram) or 0.25 µg of Anti-Mouse CD107b (LAMP-2) FITC (filled histogram). Total viable cells were used for analysis.

Description: The eBioABL-93 monoclonal antibody reacts with mouse CD107b, also known as lysosomal-associated membrane protein-2 (LAMP-2). CD107b is a highly glycosylated protein of approximately 120 kDa that is expressed intracellularly in lysosomal membranes. It is also transiently expressed on the surface of cytolytic T cells during degranulation, but to a lesser extent than CD107a. It has been reported that CD107b may be identical to the mouse Mac-3 antigen.
Applications Reported: This eBioABL-93 (ABL-93) antibody has been reported for use in intracellular staining followed by flow cytometric analysis. It is also reported for use in surface staining in a flow cytometric based degranulation assay.
Applications Tested: This eBioABL-93 (ABL-93) antibody has been tested by intracellular staining and flow cytometric analysis of thioglycolate-elicited peritoneal exudate cells. This can be used at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b) is a member of a family of membrane glycoproteins. LAMP2 provides selectins with carbohydrate ligands, and may play a role in tumor cell metastasis, and have a function in the protection, maintenance, and adhesion of the lysosome. Prior to posttranslational modification, LAMP2 is approximately 46 kDa polypeptide. Mature, functional LAMP2 is extensively glycosylated with a variety of different N-linked and O-linked oligosaccharides with a total molecular weight of about 100-130 kDa. LAMP 2 has a structure comprised of a large amino-terminal intra-lysosomal domain, a hydrophobic trans-membrane domain, and a short carboxyl-terminal cytoplasmic tail. LAMP2 is used to monitor a very selective form of autophagy called chaperone-mediated autophagy. LAMP 2 has a structure comprised of a large amino-terminal intra-lysosomal domain, a hydrophobic trans-membrane domain, and a short carboxyl-terminal cytoplasmic tail. The LAMP protein has 3 variant forms: LAMP-2A, -2B, and 2C. LAMP2, together with CD107a/LAMP-1, is a major constituent of lysosomal membrane. The LAMP proteins are involved in lysosome biogenesis and are required for fusion of lysosomes with phagosomes, and LAMP2 is an important regulator in successful phagosomal maturation. LAMP2 deficiency causes an accumulation of autophagosomes in many tissues leading to cardiomyopathy and myopathy (Danon's disease).
