CD24 Antibody (368-0247-42) in Flow
Normal human peripheral blood cells were stained with Mouse IgG1 kappa Isotype Control, Brilliant Ultra Violet 805 (Product # 368-4714-81) (blue histogram) or CD24 Monoclonal Antibody, Brilliant Ultra Violet 805 (purple histogram). Viable cells in the granulocyte gate were used for analysis, as determined by 7-AAD (Product # 00-6993-50).

Description
The SN3 monoclonal antibody reacts with the human CD24 molecule, also known as Heat Stable Antigen (HSA). This 35-50 kDa molecule is anchored in the plasma membrane via phosphatidylinositol and is expressed by erythrocytes, thymocytes, peripheral lymphocytes and myeloid lineage. It has been reported that P-selectin (CD62P) binds to CD24.
Applications Tested
This eBioSN3 (SN3 A5-2H10) antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. This may be used at 5 µL (1.0 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
Blocking Buffers
When using two or more Super Bright, Brilliant Violet™, Brilliant Ultra Violet™, or other polymer dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer or Brilliant Stain Buffer for more information.
Light sensitivity
This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light.
Fixation
• Samples can be stored in IC Fixation Buffer (Product # 00-8222) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation.
• Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.
• Our internal testing suggests that Brilliant Ultra Violet™ 805 (BUV805) is compatible with short-term methanol-based fixation, but should not be stored in buffers containing methanol for longer than one hour.

CD24 is a 35-50 kDa glycosylphosphatidylinositol (GPI)-anchored glycoprotein expressed on a variety of cell types, including erythrocytes, thymocytes, peripheral lymphocytes, and cells of the myeloid lineage. It is variably glycosylated, resulting in heterogeneity in molecular mass across different cell lineages, which can affect antibody staining levels on lymphocyte populations. In the context of B-cell development, CD24 is expressed from the pro-B-cell stage in the bone marrow through to mature, surface Ig-positive B cells, with very low or negative expression on plasma cells. It is also present on the majority of B-lineage acute lymphoblastic leukemias, B-cell chronic lymphocytic leukemias (CCLs), and B-cell non-Hodgkin's lymphomas. CD24 is thought to play a role in regulating B-cell proliferation and maturation, as well as in controlling autoimmunity. Additionally, CD24 has been reported to interact with P-selectin (CD62P), indicating a potential role in cell adhesion and signaling. CD24 is a valuable marker for studying B-cell development, leukemia, lymphoma, and autoimmune regulation, as well as its interactions with other molecules like P-selectin.
