CGL78 encodes a small chloroplast-localized auxiliary factor (the algal homolog of Ycf54) that is associated with thylakoid/pigment biogenesis and is required for normal chlorophyll accumulation. In Chlamydomonas reinhardtii, CGL78 functions within the chlorophyll biosynthetic pathway at the oxygen-dependent Mg-protoporphyrin IX monomethyl ester cyclase step that forms the isocyclic E ring en route to protochlorophyllide; consistent with this role, reducing CGL78 expression causes chlorosis, decreases total chlorophyll, and destabilizes photosystem I-light-harvesting complex I supercomplexes with reduced accumulation of PSI subunits (including PSAK). Across oxygenic phototrophs, Ycf54/CGL78 is viewed as an auxiliary component that supports cyclase activity (e.g., by promoting the level and/or activity of the catalytic AcsF/CycI subunit), thereby coupling tetrapyrrole cofactor production to photosystem assembly and photosynthetic competence, particularly under stress conditions such as copper deficiency where CGL78 is induced.
