The CHIL3 gene encodes the protein chitinase-like 3, which is primarily secreted by macrophages and is involved in immune regulation, tissue remodeling, and inflammatory responses. CHIL3, also known as Ym1, belongs to the chitinase-like protein family but lacks chitinase activity due to mutations in its catalytic domain. It is predominantly expressed in lung tissue, where it plays a vital role in asthmatic responses and allergic inflammation. Furthermore, CHIL3 exerts anti-inflammatory roles during pathogen infection by modulating macrophage pro-inflammatory activity and promoting the Th2 immune response. Beyond immunological involvement, CHIL3 is integral to tissue repair mechanisms and cancer cell proliferation, contributing to extracellular matrix restructuring and angiogenesis. It facilitates macrophage polarization towards an M2 phenotype, which is associated with tumor progression and metastasis. Thus, CHIL3 is considered a potential biomarker for inflammatory diseases and cancer, with ongoing research examining its therapeutic modulation.
