CHLH encodes the H subunit of magnesium chelatase, a chloroplast-localized protein that functions at the interface of the chloroplast stroma and envelope membranes, where it can associate with envelope fractions in an Mg2+-dependent manner and participate in plastid-to-nucleus signaling contexts. It is a large, soluble tetrapyrrole-binding protein (approximately 140-150 kDa in photosynthetic organisms) that forms a cage-like architecture composed of a major domain containing an internal cavity plus a smaller N-terminal domain connected by a linker; conformational rearrangements and loop movements help envelop the porphyrin substrate within an active-site pocket. Biologically, CHLH is the protoporphyrin IX-binding component of the ATP-dependent magnesium chelatase complex (with partner subunits that provide motor/ATPase activity), catalyzing insertion of Mg2+ into protoporphyrin IX to generate Mg-protoporphyrin, thereby committing tetrapyrrole flux to chlorophyll biosynthesis essential for photosynthesis and chloroplast development; genetic and biochemical studies also support additional regulatory roles in guard cell abscisic acid-responsive signaling via protein-protein interactions, although this signaling function has been debated across species.
