Western blot was performed using Anti-CNOT7 Rabbit Recombinant Monoclonal Antibody (Product # 703786) and a 32 kDa band corresponding to CNOT7 was observed across the cell lines tested. Modified Whole cell extracts (1% SDS) (30 µg lysate) of Hep G2 (Lane 1), Jurkat (Lane 2), LNCaP (Lane 3), MOLT-4 (Lane 4) and THP-1 (Lane 5) were electrophoresed using Novex® NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). CNOT7 (CCR4-NOT transcription complex subunit 7) has 3'-5' poly(A) exoribonuclease activity for synthetic poly(A) RNA substrate. Its function seems to be partially redundant with that of CNOT8. CNOT7 is a catalytic component of the CCR4-NOT complex, which is one of the major cellular mRNA deadenylases, and is linked to various cellular processes including bulk mRNA degradation, miRNA-mediated repression, translational repression during translational initiation, and general transcription regulation. During miRNA-mediated repression, the complex seems to act as a transcriptional repressor during translational initiation, as well. Additional complex functions may be a consequence of its influence on mRNA expression. CNOT7 associates with members of the BTG family such as TOB1 and BTG2 and is required for their anti-proliferative activity.
