CONSTANS encodes a nuclear, plant-specific zinc-binding transcriptional regulator expressed predominantly in leaf phloem companion cells, where it integrates photoperiod and circadian timing cues to control the floral transition. The protein is modular, with tandem N-terminal B-box-type zinc finger domains that support protein-protein interactions and higher-order oligomerization, and a C-terminal CCT (CONSTANS, CONSTANS-like and TOC1) domain that contributes to nuclear localization and transcriptional regulatory activity. Under inductive long-day conditions, CONSTANS activity promotes flowering primarily by activating transcription of FLOWERING LOCUS T in leaves, thereby increasing production of the mobile florigenic signal that triggers reproductive development at the shoot apex; biochemical studies also support direct recruitment of CONSTANS to cis-regulatory DNA elements in the FLOWERING LOCUS T promoter via mechanisms requiring the CCT domain.
