Total proteins were isolated from 7-day old Arabidopsis thaliana Col-0 (wild-type) and coi 1 (null mutant) seedlings according to [ref 1]. COI1 encodes a predominantly nucleus-enriched F-box protein that functions as the substrate-recognition component of an SCF(COI1) E3 ubiquitin ligase complex in plant cells. The protein is modular, with an N-terminal degenerate F-box motif that mediates association with SKP1-like adaptors (enabling assembly with CULLIN1 and RBX1 into an SCF complex) and a C-terminal leucine-rich repeat domain that forms a ligand-binding pocket and provides surfaces for partner recognition. Biologically, COI1 is essential for jasmonate signaling: together with a JAZ transcriptional repressor, it forms a hormone-dependent co-receptor that binds the bioactive conjugate jasmonoyl-L-isoleucine and is potentiated by an inositol pentakisphosphate cofactor, thereby promoting recruitment, polyubiquitylation, and 26S proteasome-mediated degradation of JAZ repressors to activate jasmonate-responsive transcriptional programs. Through this mechanism, COI1 controls diverse jasmonate-regulated outputs, including wound- and herbivory-induced defenses, pathogen-related responses, growth modulation, and reproductive development (notably fertility).
